Everything you've read in previous answers is true, but something was left out that I thought might interest you. Gwen Stacey will be a 'love interest' of Parker's, however the reason he is unfaithful to Mary Jane is due to his new suit. It's a symbiotic lifeform (probably created by Ozcorp although the comic books details it a lot differently) that makes a neat double as a suit because it adds strength and bulletproofness. But it also beings out the dark side in Peter Parker. So although there will be a romance between Parker and Stacey, it will likely be more as a result of Peter's new evil side and nothing to do with true love on his part. It's also rumored that James Franco will become some rendition of the Hobgoblin, which probably means that he'll kill Gwen Stacey. Even more exciting is that the black suit and Parker eventually parted, and the suit found a new host- Eddie Brock, (played by Topher what'shisname?) and became venom. Venom being my favorite villain in the Spidey series, I am extremely excited about this movie. You can find jpeg's of what is supposed to be the venom from spiderman 3 all over the Internet right now. it could be fake, though...

Gwen Stacy will be in the third movie. The actress, Bryce Dallas Howard, has been cast for the role. There will be a love triangle that develops between Peter Parker, Mary Jane and Gwen Stacy. In the comics, Gwen Stacy is Peter Parker's girlfriend who dies when the Green Goblin throws her off a bridge. However, the movie plotline will definitely be different since they have filmed a similar scene in the first Spider-Man movie.

A blonde, bookish girl, Gwen was the first true love of the nerdy Peter Parker (Spider-Man). His adversary the Green Goblin murdered Gwen in the The Amazing Spider-Man #121 (June 1973). Both the decision to kill Gwen and the method in which Marvel implemented it are controversial among fans, but it is still a pivotal point in both Spider-Man’s history and in American comic books in general. Spider-Man writers and fans disagree about who is the character’s “one true love,” Gwen or his subsequent wife Mary Jane Watson.
