Are you talking about a stand-alone, or a series, and then, what genre?
As far as series, if you mean romantic comedy- Maison Ikkoku hands down (14 volumes)!
Of Tom Clancy-like political machinations- EAGLE: The Makings of an Asian-American President (4 volumes @ 19.95 ea)
For SF- Akira (I know this is at 4 volumes, 300-plus pages, but if you read just the first one, which is all I've read so far, it's so incredible in plot and detail. Blows one's mind more than even its indelible anime version)!
With a stand-alone, there are countless short-story, single volume collections for many tastes:
Short Program and Short Program 2 by (last name first) Adachi Mitsuru (He's the master of the manga short story).
His stories are true-to-life, yet he makes the most mundane things exciting, and his endings are a surprise.
When *I* think about the best manga book, I think in series, or do you mean in a single volume short story collection? There are few manga that tell a story in one book.

Personally,
I think the best one's that I read at this point would be :
'Blade of the Immortal' by Hirokai Samura, 'Vagabond' by Takehiko Inoue, and 'Neon Genesis Evangelion' by Yoshiyuki Sadamoto....not to mention the classic 'Akira.'
All drawn with intelligent artwork (classic, non-heart shaped faces) and more mature intelligent storylines than the majority of U.S. released manga out there.

Its just Manga, dont put book after it and there is no one best, there are many, such as Bleach, Fruits Basket, Full Metal Panic, From Far Away, and so on.
