Closing down the flap on the vent that lets cold air into the room AND closing the door will make a small difference, as long as you don't do that to the room with the thermostat.

u can close the vents and force more air to other rooms, but if u close off a room make sure that room doesnt have a return in it. if a return is in a unconditioned room u will be forcing warmer air into the system and causing it to work more! so close off the return also!
plus, if no return in the room, make sure that u seal the door with a towel if there is a large gap under the door. again, u will be pulling hot air from under the door of the unconditioned room!
