I really like Tom Hanks and Meg Ryan, but my favorite on screen couple would have to be Adam Sandler and Drew Barrymore. They were so good together in "The Wedding Singer" and "50 First Dates." It is a toss up on which on is better. They seem to play of each other so well, to the point that it looks as if they are just being themselves. They make a great couple. Sandler is a lucky guy.

As far as male and female couples I'd say Kurt Russell and Goldie Hawn and of course that has carried on beyond the screen as well.
Old school comedy..... Burt Reynolds and Dom Deluise, Don Knotts and Tim Conway, Mel Gibson and Danny Glover
female actresses: I know its television and not the movies but I have to go with Penny Marshall and Cindy Williams (Laverne and Shirley) They played off each other perfectly.

I liked Rupert Grint and Emma Watson in HP & Goblet of Fire and I loved Brad Pitt and Angelina Jolie in Mr. & Mrs. Smith. Julia Roberts and Richard Gere in Pretty Woman was also fantastic.
