I'd have to say Land Rover Safari, but followed closely by Toyota Land Cruiser, a vehicle I owned at one time. Both offer the necessary gear ratios and number of forward and reverse gears to make them unbeatable off the beaten track. The British Land Rover is used extensively in the African bush country, while the Japanese branded Land Cruiser has been a favorite in South America and its rain forests for decades.

By excluding flying, are you excluding hovercraft?
If not, then some other small amphibious vehicle, such as an Argocat (http://www.argocat.com/ )
If you're only talking land-based off-roading only, then the LandRover Defender is good (when its working properly), the old Suzuki SJ413 is pretty crude, but effective (its light weight and small size are big advantages at times - they're great for off-road trials), and the Pinzgauer (http://www.automotive-technik.com/ ) and Unimog (http://www2.mercedes-benz.co.uk/content/unitedkingdom/mpc/mpc_unitedkingdom_website/en/home_mpc/unimog.html ) are very effective.
However, the Argocat might still be better.
Or a small hovercraft.

The best one is the one you can afford and the one that YOU have fun with. You can buy an offroad vehicle for $1,000,000, but that doesn't mean it will be the best.

Only In a Jeep!!! Jeep all the way baby!! They run forever! Plus they were the 1st 4x4 made so I trust them over all the 4x4 makes N models.

CVR(T), British built personnel carrier, can float on water(using special skirts) and will never sink in poor ground due to it's low pressure foot print.
best thing it does 50mph off-road forwards AND backwards
