Assuming your e-mail program is open, Windows has a way to set a sound to occur when new mail comes in. To do this:
1. Go to "Start", then "Control Panel", and then double click on "Sounds and Audio Devices".
2. Click on the tab on top that says "Sounds".
3. Scroll Down and click on "New Mail Notification".
4. In the space for sound, enter the path by clicking on the Browse button and choosing a sound, usually found in the C:\Windows\Media folder.
5. Then click on "Apply", and then "OK".

Then whenever new mail comes in while the mail program is active, you'll hear the sound you chose. Good luck.
