Closing the vent to outside air re-circulates the air inside the room. Arguably by re-circulating the air you will cool the room more quickly. This may be true depending upon the temperature of the outside air and the ambient humidity level.

If the room is cooler than the outside, close the fresh air vent. If its cooler outside, open the fresh air vent. You should open the vent once in a while to freshen the air though.
