The key to running fast 40 times is to have strong, explosive hip flexors. When you do lower-body weight training, incorporate explosive moves, like weighted box jumps, power cleans, etc. Make sure you work your hip flexors though, and practice running 40's. Practice running 50 yard sprints and practice "getting out of the box".

When you work out your legs, Don't use heavy weights. Do light weights and more reps, and do them quickly. Don't do squats, do more explosive type exercises(calf raises, ladder drills ect.).
This is gonna sound funny, but run downhill. Training your legs to run faster before you can actually run faster will help your control. I would say find a slightly inclined hill, with no more than a 45 degree slope. Sprint down it, walk/jog back up.
Also, make sure when you run that your arms don't cross your body. Pump them straight up and down. When you cross your body, you waste movement.

plyometrics and weight lifting. you should probably work on exploding from a stationary position, so maybe olympic lifts?
