Generally, you would need to go to the City Hall or Village Hall of the place where you are locating the business. You will need to fill in some sort of application and pay a license fee. The application and cost vary greatly from city to city, so it would be difficult to guess what it would be in your locale. Remember to submit paperwork to your federal and state agencies, too, to get yourself set up for sales tax (that should be filed with the state only and possibly the city, if your city charges a special sales tax), and business income and payroll taxes (both federal and state on that one). If you haven't retained an accountant, now would be a good time to do so, just to make sure you're heading down the right path.
