Even if the Interstate speed limits are 70 or 75 MPH, I always plan my long distance trips for about 60 MPH, to account for fuel, food and rest stops (summertime highway construction zones too). At 60 MPH it would take you about 10 hours to go 600 miles.
If you leave Colorado Springs at around 8AM, you should be near Omaha, Nebraska at 6PM. Probably as good a place to stop for the night as any. Another 10-hour day will get you to K'zoo.

If you drove the 600 miles at exactly 70 mph the entire way it would take 8 hours 34 minutes and 17 seconds. So, plan on about 8 1/2 hours.

I regularly drive from Denver to Chicago, and it takes about 16 hours of driving time. Add 100+ miles or about 2 hours from Chicago to Kalamazoo and 1 hour from Co Springs to Denver, and total driving time will be 19 hours which divided by 2 gives you 9.5 hours. There are plenty of motels along the way - pick any once you reached half-way point.

I once drove from Denver to Chicago, it took about 16 hours. Kalamazoo is another 2+ hours from that.
Halfway on I-80 would be somewhere in Iowa.

Well, it will take about 18 hours... and... if you want to stop, about 730 miles into your trip, you can stop in Des Moines Iowa and that is about half way...then you just hop back on I80 east and head the rest of the way. I do a lot of road traveling so ask if you have any questions!

if you can hold a constant 70mph then about 8½ hours
but you won't be able to do that because of time lost for acceleration/braking, traffic jams, roadworks, toilet breaks etc
realistically look at 12 hours for the 600 miles

According to mapquest it will take 17 hours and 50 minutes. Why don't you stay overnight in Omaha?
Here are the directions from mapquest.
