Each place approaches perfection, depending on the taste of the visitor!
There are thousands, literally, of places which miss nothing! They are perfect! Thus, it depends on YOUR taste!
You like mountain with forests? Most likely islands with hot and dry climate, warm, blue, clean, clear skies and sea? You like to combine with learning history (museums and antiquities?), etc.
Everywhere there is great food and gentle people to meet, talk, associate with!

Gosh, that's a tough one. If I were sending tourists to Greece, and they could only go to one place after a day or so in Athens, I would send them to Santorini. It's incredible. Unbelievable beauty! However, there are sooooo many other beautiful places, some I like more than Santorini. But Santorini is the most impressive on first sight and offers the best tourist bet.

This question is impossible. Even tiny little back water villages
on some remote Greek Isle can be very enticing. Many of
the islands are desolate(very few trees) and the plumbing is not
always the greatest, but that being said, Greece is a fun place
to be 100% of the time. You don't ever get tired of visiting or
even living there.

The Isle of Paros. Arrive at the port and take a bus to the far side of the island (maybe 20 minutes). Camp on the beach. Crystal blue waters, slight breeze, little grocery store down the way. Not certain why I left exactly...
Did I mention it was a nude beach? Didn't realize that until we woke up the next morning. So we stayed a few extra days - HA!

If you want sea you should visit Naxos, Santorini, Crete, Xalkidiki.If you prefer the mountain you should visit the lake named Plastira, Meteora. There are a lot of beautiful places in Greece!!

The sunset from the cliffs of Oia on the island of Santorini. If you plan on staying in Santorini don't do it in Fira, instead stay in on of the villas on the cliff side facing the sunset in Oia, if you want to know what it is like go rent Summer Lovers (1980's movie with Daryl Hannah and Peter Gallagher), the villa they stayed in, in the movie is gift shop on the cliff side in Oia. Also you don't have to fight the traffic with everyone coming from Fira and then packing in up at the old church ruins, instead enjoy the sunset from your own balcony.

It depends of what you want!.
Sea or mountain?.
The most beautiful island for me is Santorini, and the most beautiful town is Athens,or you could go to Olympos mountain for winter, the mountain of ancient Gods!.
Summer or winter Greece has everything to see!.

If I have to choose ONE place then it has to be Santorini (of course on summer).
But I really believe u should give more details like if u really like beaches then I believe the bests are in Milos, Nightlife -> Mykonos or Athens, touring. site seeing -> Crete.
