Well, since you've had the dream your whole life, you'd start out with, "It has been my lifelong dream ..."
It also depends what you mean by "course". If you mean a particular plan of action, you might say,
"It has been my lifelong dream to pursue this course," or "...to follow this course."
If you mean "course" as in a school class, you'd say, "It has been my lifelong dream to take this course."
If you're in a restaurant, you'd say, "It has been my lifelong dream to eat this main course, followed by dessert." (Yes, now I'm just being silly.)
