If you use Yahoo e-mail, you will only get an alert if you have Yahoo Messenger running.

The speaker in your computer should beep when you get a new email.

Hook up a circuit to detect that, and have it trigger a photoflash on top of your monitor.

You need to include your email program when posting a question such as this. As the list for all email programs would be prohibitive.
