if you had your first interview and the person interviewing you gave you conditions you didn't know before and aren't flexible with, how do you say I don't think I want to work here anymore at the second interview with the manager?

You should not feel obligated AT ALL to take a position which makes you uncomfortable. Remember, the interview process works BOTH ways...you are interviewing them as well! If you're new to the process, take heart, there are more companies out there and better opportunities. If I was you, I would simply say that I'm looking for a different kind of opportunity than the one they're offering...and leave it at that. Unless this is the ONLY job in town...or it's a relative/friend...you have options, exercise them!

I would just tell them why you are unable to meet the conditions. If they like you enough, they may decide to be flexible on their conditions, especially if you have a good reason. Just make sure that you get any exemptions in writing so they cannot come back later and demand you meet their conditions.
