Make sure that you have everything lined up with TDC. #1 spark plug should be all the way up on the compression stroke and the rotor inside the distributor should be pointing at the position of the #1 wire in the tower, Then you install the balancer on TDC or 0. As long as you have everything on TDC the engine should fire rght up from there.

Just in case you need to know it, to make sure that you're on the compression stroke: pull #1 spark plug and have a Friend BUMP your starter while you press your finger over the spark plug hole. when the motor trys to push your finger off, stop using the starter and turn the balancer in the same direction until you reach TDC. Just in case you needed to know!

If I understand you Q , the answer is no. once you have the engine built and in time, you can start it from any location on the rotation.
