Yes, Divine was actually a man. If you watch Hairspray, he played dual roles as Tracy's mom Edna Turnblad and the bigoted station manager Arvin Hodgepile.

To start with Divine is dead and has been for quite awhile.
Next Divine was male his real name was Harris Glenn Milstead and he was nicknamed "Divine". He was a cross dresser. He came from Baltimore and was a friend of John Waters who of course featured him in "Pink Flamingos", "Female Trouble", "Polyester", "Hairspray" and a couple of other movies.

Divine is a man dressed in drag. His real name was Harris Glen Milstead. He met director, John Waters, when they were in high school together and made a series of low budget, but very good, comedies that launched John Waters in fame.
