You can create an OleDb connection string for an Excel file by doing something like this:

string ConnStr = "Provider=" +
"Microsoft.Jet.OLEDB.4.0;Data"
+ " Source=" + ExcelFilePath +
";Extended " +
"Properties=\"Excel " +
"8.0;HDR=Yes;IMEX=2\"";

....where "ExcelFilePath" is the physical path to the file. Then just populate a DataTable like you would with any other ado.net data source.
