The shortest title reign was Andre the Giant. He held the belt for 45 seconds after defeating Hulk Hogan, but then he tried to sell it to the Million Dollar Man, which prompted the WWF to vacate the title.
The second shortest was probably Yokuzuna, who won it from Bret Hart at Wrestlemania, but then lost it minutes later to Hulk Hogan.

Eddie Guerrero, i think, haven't watched wrestling in a long long time.
*Edit
I've just noticed this question can be taken two different ways, by shortest do you mean the length of time the title was held or the actual size of the wrestler?

yokozuna beat bret hart at wrestlemania 9, then right after that match ended Hulk Hogan came out and beat yokozuna (due to mr. fuji accidentally throwing salt in yoko's eyes) to become the new champion, much the same way edge came out to beat john cena when he cashed in his money in the bank.
