A LOT.

Scarface (obviously, the epitome of drug movies, at least to many), Traffice, Fear and Loathing in Las Vegas, Blow, Requiem for a Dream, Thirteen, Sid and Nancy, Boogie Nights, Dazed and Confused, and many more. Here's a list of movies with drugs, etc: http://en.wikipedia.org/wiki/Drug_movies

I dont know if you could say the film is souly based on drugs but Pulp Fiction has a HUGE segment on herion and its an amazing film.
Also Traffic is about drugs which was also quite good.
And also "Go" was soo good it was about ecstacy and its really cool to watch.
