My favorite is a scene from "I'm Gonna Git You Sucka" (I saw it when I was a kid), where this guy is about to get lucky with this hot chick, but she has some hidden secrets.
I can't remember what comes off first, but she is wearing a wig because she only has a few strands of her own hair, I think she has fake teeth, she has a fake butt that falls off. I think her boobs fall off too and her fake leg falls off. It sounds horrible, but it really is the funniest thing I have ever seen.
Oh, I completely forgot about Monty Python and the Holy Grail. The scene with the Black knight when he gets his arm chopped off, and Says,
BLACK KNIGHT:
'Tis but a scratch.
ARTHUR:
A scratch? Your arm's off!
BLACK KNIGHT:
No, it isn't.
ARTHUR:
Well, what's that, then?
BLACK KNIGHT:
I've had worse.
ARTHUR:
You liar!
BLACK KNIGHT:
Come on, you pansy!

To be "the funniest" I think it also has to be the most "durable"..... you can watch it a million times and it's humor doesn't diminish through the decades.
That leaves most sight-gags and stunts out.
Jokey scenes lose their durability with heavy viewing.
That leaves a good character-developed scene built upon the whole movie and encapsulated into a short-term experience.....
We're talking a Bill Murray or Jack Nicholson kinda' film where any part of it is like a hologram.......take a piece out and it reflects the whole thing.
Take it even further.....remove all other characters from the scene........we are distilling the ESSENCE of the humor/irony/pathos into a scene with one star, a set and a camera.
We've got Bill Murray in "Groundhog Day"
and Jack Nicholson in "As Good As It Gets".
It's so hard to choose between Bill trying to commit suicide 1,000 ways and Jack trying to deal with a little poochy-woochy.
But to give Jack the credit that he is TRULY due......after SO MANY unbelievable performances ........ let's go with him throwing the a peeing dog down the
garbage chute !!!!!

My personal favorite comes from "You Only Live Twice".
At the climactic crater battle, the bad guys are holed up in a seemingly impregnable bunker.
The good guys are shelling it to gain entrance.
Ernst Blofeld (played by Donald Pelasance) is petting his cat.
When the explosions go off, Donald is holding the cat firmly by the scruff of the neck. Meanwhile, the cat is totally and absolutely freaking out from all the noises and is proceeding to dig her claws into Donald's arm in a vain attempt to escape.
Later on, you see Donald holding his arm in pain as the cat did some major damage.
Next time it comes on, check it out.
I have never laughed so hard in my life, "Airplane" included.

Definately the scene from Bruce Almighty where he makes the newscaster say gibberish on camera. I laugh so hard I actually cry.

In the movie Waiting with Jason Long and Ryan Reynolds...throughout the entire film there is this one guy who is ignored by everyone and at the end there is a party that they all go to after work. Well that one guy stands up on this table and shouts out all these things that he hates about each one of the people who has ignored him...then he points to comedian Dane Cook and just points at him and says" and YOU!!!!", the funny thing is that they have never even met. Then the guy just pulls down his pants and shows everyone his balls. It was a game played throughout the entire movie and he just shouted at Dane Cook to get everyone's attention...IT'S HILARIOUS and if you haven't seen it YOU SHOULD!!!!!♥

The newcaster in Bruce Almighty was Pretty funny. But I really enjoyed every scene in Van Wilder.

The Black Knight scene from Monty Python and the Holy Grail wins the award. And the runner-up for me is the vomitting scene from Team America World Police. You can never really appreciate the humour of someone else throwing up until you see a puppet throwing up.

To this day and call me a Star Wars nerd if you want
but I always loved in Empire Strikes back when they are tyring to fix the hyperdrive and c3p0 says that there's possible a problem with some cuppling, and Han acts like he doesnt believe him and then turns around and tells chewie they need to replace the cuppling and c3p0 gives him a look

well from "fun with Dick and Jane", the funniest scene is where they are in that big guys house stealing a painting and they have ski masks on. Then Jim carrey keeps making these funny noises and he has a voice changer on, and I could not stop laughing. When I saw it in the theater, people were literally rolling around on the floor!

OMG >>>> it has to be in the movie "Wedding Crashers" when Owen and Vince are hunting and Owen fires the shot gun and goes plowing into the ground while Vince is getting shot in the @ss by the buttheadd boyfriend. Or, it could have been when he was getting the handjob under the table too. You make the call....
