In 'Outlook' highlight the sender to be blocked, click 'Message'/'Block Sender'. Alternately, 'Tools'/'Message Rules'; and continue from there.

If you use Yahoo! mail, whenever you receive an unwanted E-mail, click the little checkbox next to the E-mail, then click the "Spam" button at the top of the page. After 2 or 3 times of doing this, the E-mails will end up in your "Bulk" aka "Spam" folder.

depending on the email provider it is easy to block emails. For hotmail go to options and then click the mail tab. When the page loads click block senders. This then allows you to enter the email address of the person you want to block.
By doing this no emails will come to you, and any email they try and send will be returned as undelivered.

good answers but you can always set up a filter so the emails go straight to trash if it's someone you know an ex perhaps, reporting the email as spam reports that user as a spammer and the details are updated to the spam filter that gets forwarded to everyone with that spam filter.
so if the email is from a known source that you no longer wish to comunicate with your first choice is to ask them to stop sending you emails including a do not repsond to this email with the stop request.
you then set up your filter with your email provider most email clients/ accounts are able to set up filters. see the help section for your email client (yahoo, msn etc.)
once the filter is in place check your trash can daily for messages from that user. What i forgot to say is keep a copy of the emails (so set up a new folder called nuisance and keep a copy of the emails from that person in there and a copy of your stop request) if after sending the stop request you still get emails from the annoyer then you can take it up with the company hosting his email account support or customerservice@companyname.com as an example. forward them copies of the emails you have recieved and a copy of your stop request, they should be able to block that user from sending you email, unless that person creates a new email address then you will have to repeat the process again.
once thats done give it upto a month for the emails to stop. If they don't then you should have a case for legal action seek proffessional advice.
only report genuine spam as spam because as i said this will block that account on all other systems using the same spam engine as yours.
