Since allosteric enzymes use an alternate binding site for the regulatory molecule, changing substrate concentration does not generally alter the effect of the enzyme's allosteric properties. However, increasing the concentration of the substrate could potentially make the substrate binding site less accessible (simply because it would always be occupied)

things might not change at all depending on the enzyme, but it's dependent on the type of enzyme and if inhibitor is present etc.
