For strengths, if you're not sure, check with friends and colleagues on what they think your strengths are. If you're going to a job interview, highlight your strengths that will benefit the potential employer, based on the skills and needs they have for the person they're hiring.
For weaknesses, I agree with the last response. You need to be honest. You should also state what you are doing to improve your weakness. This shows initiative and growth. I would only highlight one or two weaknesses, and they should not be something that would prevent you from getting the job you're interviewing for. For example, if you're interviewing for a bookkeeping job, saying that you're not good with numbers is probably not a good idea.

I assume you mean during an interview...just make sure your weakness is a genuine weakness - not, for example, being a perfectionist or working too hard.
