This way each audience member would fill in the blank with their own ultimate contents. All you were supposed to know was that it was "so beautiful." No prop master can come up with something better than each individual's imagination. At least that was the original idea. Then somebody had the bright idea (which I think was a mistake) of putting an orange lightbulb in there. Suddenly what could have been anything became anything supernatural. Didn't need to push the effect. People would have debated it for years anyway, and it would have been much more subtle. I can't believe I'm actually talking about being subtle."

The briefcase contained the soul of Marsellus Wallace. When the devil takes your soul, he takes it from the back of the head, hence the band-aid. The briefcase contained his soul, the three guys who had it worked for the devil and that's why the combination was 666. The "divine intervention" actually was, God was helping them save a soul.

It was meant to be Marsellus Wallace's soul that was stolen from him at the beginning (note the band-aid on his neck). While others would speculate that it could be whatever you want it to be (as mentioned in the DVD extras), the original idea was the theft of Wallace's soul and carry it around in a huge, shiny briefcase.

I think the answer I have heard that makes the most sense is that it is his soul. That explains why when they get shot at directly, the bullets don't even touch them. They are saving a soul from hell, and that is a good thing.
But having no explanation makes it work too-making it anything you want it to be allows you to construct your own explanation, as long as it works with the other events in the movie.

A possible explanation for what was in the case was that it was something metaphoric, conceptual... like "happiness" or "salvation" or whatever number of things that you can think of that most EVERYONE wants and would do almost anything for.
