Keep in mind I am a teacher and will be travelling with 2 other teachers and about 20 high school students.

The Palace on the hill is really cool. It is the old walled city and has cool shops in the wall, armories, the cathederal. Actually the best Midievel Castle I have yet to see.

John Hus square is interesting, has the astronomical clock. Near there is the Bethlehem Chapel which was John Hus's Church. I don't know if you have heard much about him. He was a priest why stood up against the selling of indulgences and was sent to the council of Constance and burned at the stake. Something of a patron saint of Prague.

Of course you should take them to the st. Charles Bridge where they filmed Mission Impossible. The Opera house is there too. I think near the Palace there is also an old Jewish quarter. Beautiful city. Try and take in a concert opera if you can as well. And watch out for gypsies and pick pockets.

I recommend the Museum of Communism. It takes about 2 hours to go all the way through it but there are some fascinating artifacts, sets & information. The teachers will especially like it as they grew up in the cold war era. The have some heavy propaganda against the "evil" capitalists. Anyway, very interesting.

You have to see the Castle. You'll want to check out Kafka's old apartment. You'll have to go to Old Town and see the Astronomical Clock and everything else. Check out the Jewish quarter. Everything there is interesting. There's a lot to do.
