I have a PA-b6350 soundboard/amp, and two speakers.

The amp has two 1/4 inch output for speakers. the speakers have 2 1/4 inch inputs.

I want to run some speaker wire to the speakers. When I wire them up, do I use a 1 "stereo" jack to speaker wire on the amp, and 2 "mono" jacks (one on positive, one on negative) for each speaker?

You need to check with the manufacturer or a stockist of the loudspeaker exactly what the two jacks are for.

The need for a stereo jack plug at the amplifier end is highly unlikely.
