A query letter is a letter you send out when you are trying to find out if an agent is willing to represent your book. Usually it includes a BRIEF (2-3 sentences) about yourself and a hook for your manuscript. With the query letter only include the first 3 chapters of your book. This is so the agent can see your writing style and see if you are any good at writing and if the story is intriguing. Also it is good to include a 1 page plot summary. (This summary would be written in the present tense).
There are tons of resources for writing query letters online and in books. The Writers Market is a great book for publishing in general. It lists agents and publishers and what types of books they represent. They also have some helpful hints on how to write query and cover letters. Online a great website is www.preditorsandeditors.com

It's a letter asking for a sale or asking about the possibility of a job. It is used frequently when trying to sell story manuscripts to publishers. The letter goes along with the work and introduces the creator of the work and asks that the publisher review the work with an eye to possibly buying it from the creator.
