You're providing an air-tight seal between the slide and cover slide that reduces diffraction through several layers (air, liquid, sample surface, slide backing). It also reduces thermal perturbations from heating the sample via indirect lighting of the sample.

To straighten the light waves so that you can see at higher magnification. With that lens you can see upto 1000 times magnification. Yo can use other things instead of oil e.g. Xylene which I use quite often
