you question is pretty broad, it depends on the "something" you want removed. you can prevent it from running by following these steps
1. Click Start
2. Click Run
3. type msconfig.exe
a system configuration utilities dialog box should come up
4. click on Services
you will see a serious of services running on your system. and then click on the box (basically unchecking) the box that you don't want the service to run. And click Apply and Ok. This may require a reboot.
Also you can take these steps
1. Right click on My Computer icon
2. Click on Management
3. Expand (click on the plus sign) on Services and Applications
4. click on services
5. click on the service that you want to stop running.
note: by doing it this way, it gives you a description of what the service does for your computer, when you click on the individual services. I would suggest doing it this way so you are not unwittingly stopping any services that are critically to your computer successfully running.

Get a really big pair of tweezers. Or a wrench. If that doesn't work, take a sledgehammer to it.

If you want to remove it permanently, you have to either unistall the program or remove its registry entry from the following keys:
HKLM\Software\Microsoft\Windows\currentversion\run and
HKLM\Software\Microsoft\Windows\currentversion\runonce
