first of all spend a lot of time drawing the figure from nature, learning form, gesture, balance , anatomy, fore shortening and proportion. Then once you are reasonably skilled at that I would try sketch some heads from photo reference. then I get some tracing paper and make several variation of my sketch. Do several traces over a trace each time getting a little more stylized. Feel free to look up anime styles you like and try to emulate them. A good exercise to try is do a caricature(in magna style) of someone famous, someone instantly recognizable that everyone has a preconceived notion of how they look.See how far you can stretch a caricature and have it still read as the celebrity. the better you can draw figure the better can apply that skill to exaggerating the figure. the better you can copy nature, the better you can distort nature to suit you needs.
There are however no easy answers and no short cuts. Spend a lot of time, use a lot of tracing paper. and I would do alot of gestures.

After or with sketching on papers or tracing, you can also use Adobe Photoshop or Illustrator for drawing your characters. You can scan your sketch from paper or tracing and then color over it and easily give a story sequence like look.
