depends on the operating system of the computer you're talking about.
For older versions of windows (prior to XP) you can buy software to do this for you, but there's nothing built in. ('Protect Folder' is the name of one such application)
If you're running Windows XP, you can do this using the technique of making the folder compressed, and then encrypting the compressed file for the folder. Details of the process are explained here:

Well it isnt possible to secure a folder by passord...only thing i have ever known was right click the folder and go to properties and go to sharing and make the folder private from other users names on the computer or you can open the folder and go to the TOOLS tab and click on folder options and then click on view and make sure it says hide hidden files and folders and then hide your folder. Thats all i can think of.

if you are using windows 98 then yes
just go in to dos view
and create a folder by a name aa (u can create it on windows explorer too)
now change this folder name with a comand now only in dos
ren space aa space (press alt button and then press 2 three times) and now press enter
now try to open that folder in window explorer
else in xp you can change the properties of folder as hidden but it is risky dear
first option is 100% secure

Here is 1 way by which u can ensure Privacy to ur folders.
Suppose u want to secure a folder named bk saved in c:.
1)Right click on folder bk.
2)Rename it to bk.{20D04FE0-3AEA-1069-A2D8-08002B30309D}
Note:- there is a dot after bk in above step2.
3) Press enter
Bang ur folder has been transformed to My comp.
Now to access that folder just go to dos prompt and rename that
folder.
eg in this case:-C:/ rename bk.{20D04FE0-3AEA-1069-A2D8-08002B30309D} bk
Now check ur folder again it 'll show you normal and click on it to
open.
