Steamboat Willie.

This was the first Mickey Mouse cartoon released in theaters, and the first cartoon with synchronized sound. As a mischievous deckhand on a riverboat, Mickey, to Minnie's delight, plays "Turkey in the Straw" utilizing an animal menagerie as his instruments. However, the tyrannical Captain Pete is not amused, and Mickey ends up peeling potatoes in the galley.

Totally awesome. Totally original.

Based on your tastes, I'd say King of the Hill.

Personally, I'm going Transformers, GI Joe, and King of the Hill in a tossup.

PS I can't believe you included American Dad on your list....it's only midly funny.

My all-time favorite from when I was a little kid, then in school and even now is Tom and Jerry.
I was just watching a show while having dinner and almost split my sides laughing!
