You actually need a few more parameters in your question. For starters, what are the dimensions of the roof? Also, What area do you live in? Climate has a lot to do with this question. For example, Colorado would require much stronger framing lumber (snow load) than Florida, (no snow load). Would this flat roof be sloped to drains in center or pitched slightly to gutters? Any equipment to be mounted to this roof? What type of roofing/moisture protection is to be applied to this roof? Is there heated living space below this roof? Or is this just a carport type roof? Is this roof going on an existing home such as adding a second story to a home? If this is the case than you would need to consult an engineer to determine if existing structure could support said second story with said roof. Okay I'll stop here cuz you are probably overwhelmed with questions and the idea: "I thought this was going to be so simple, but now I'm thinking is it worth the trouble"? not sure what your asking, but if your just talking about the roof. it depends on if the under side will be open if not 1/2 inch o.s.b board or 1/2- 5/8 plywood wood be enough they sell clips that go in between. if it is going to be exposed to view 3/4 1x8 or 1x10 would work fine about 1/4 apart for expansion. hope this help if not i most of misunder stood the question
