shemale refers to transwomen (male-to-female transgender or transsexual people) who have breasts typical of an adult female (through hormone replacement therapy and/or through breast augmentation), and who usually have other female secondary sex characteristics, but who have not undergone genital reassignment surgery.
gay is a word usually used, as either a noun or adjective, to refer to homosexuals; persons sexually oriented toward members of their own gender.
"Gay", when used as an adjective, sometimes describes traits associated with both homosexual men and women, their culture, or perceived lifestyle.

A shemale, I think, is a man who has been surgically altered to look like a woman, except he still has a penis.
A gay person is simply a man or a woman who is sexually attracted to the same sex.

Let's start at the top.
A gay person is one who is sexually attracted to persons of the same gender. (Basically, it's a boy who likes boys or a girl who likes girls.)
Shemale is a term out of the porn industry. Proper term is transsexual. A transsexual is someone who identifies with a gender not the one s/he was born with. (It's a boy who wants to be a girl or a girl who wants to be a boy.)
There is a wide variety on the transsexual spectrum, from boys with breasts to chicks with sticks to women whose gynecologist can't tell. (After sex reassignment surgery, it often is the case one cannot tell she was born male.)
The two categories are not mutually exclusive; a male-to-female transsexual may identify as lesbian.

A she-male is gender disphoric, that is, they are attracted to the opposite sex but consider that they, themsleves. are in a body of the wrong gender. A gay person is attracted to people of the same sex that they consider themselves to be. This, in itself, can be problamatic, as I've met two different men who wanted to have sex change operations so they could become lesbians. Go figure.
