This is a great question no one has the answer to, I think not even Tarantino. Well, I think it may be ingrained into the philosophy the movie tries to deliver onto the viewers.
In the scene with the shooting of the guys in the apartment, Jules, I think, opens the briefcase for the first time and whatever is inside is glowing. First, i thought it may be gold. But that it would be really heavy to carry around. So what could there be as a source of light? And why do they stare so amazingly at it?
The second time he opens is (one of the goofs you should look for next time you see the movie), Jules opens the briefcase for "Pumpkin" (Ringo), an orange light bulb can be seen through the mirror on the top part of the inside. - SO MAYBE IT'S A BULB! Just kidding :)
I think it is probably old Wallace's soul in that briefcase. Or maybe the future? When Jules walks away from the dirty business of killing people, he repents and stays alive. Vince, who stays behind, dies while sitting on the toilet for his very last time. Could there be some religious or moral message in that briefcase? Eziekel 25:17, anyone?

it's had to say, each time they opened the briefcase an ominous gold glow would happen...i would rule out gold because it's to heavy and the way they carried it around all the time it didin't appear weighted down. if i were a betting man, i'd probably have to guess it was some sort of extravagent jewerel piece.
