The glowing gold object in the briefcase is called, according to director Quentin Tarantino, a "McGuffin", a plot device used to advance the story without being revealed for what it is. The point is, you dont need to know whats in the briefcase, only that Travolta and Jackson need to get it from the 4 punks.
There have been rumors, the gold thing in the briefcase is Marsalis (Ving Rhames) soul, since in one scene you see Rhames from behind and he has a band-aid on the back of his neck, the traditional place the Devil pulls your soul from. And also, the code on the briefcase is "666", the Mark of the Beast. But the director says its nothing, dont worry about it.

IF YOU ARE TALKING ABOUT THE BAG BUTCH'BRUCE WILLIS' HAD,IT WAS`THE MONEY FROM THE FIGHT,HE THREW THE MATCH AND WASNOT SUPPOSED TO HAVE IT.IF YOU MEAN THE BRIEFCASE THAT VINCENT'JOHN TRAVOLTA'OPENS,IT WAS EITHER MONEY FROM THE DRUG SALES OR THE BALANCE OF DRUGS AND THE MONEY,THEY WERE GOING TO KEEP IT,BUT THAT DIDN'T TURN OUT TO WELL.
