The Space Shuttle program is due to be junked in 2010 - hopefully, by that time the space station will be finished. Next up is the Moon although if we don't get our act together and get around to it sooner, we'll probably meet the Chinese there since they plan on sending a manned flight to the moon as well. After that, on to Mars - and let's see what else and who else - is out there.

From what I understand, the Shuttle is about ready to be scrapped - they were high-tech in the late 70's and early 80's, but are basically antiques at this point.
Remember after that last on burned up on re-entry...they kinda reached the point of discussing new designs...?
They'll probably look sort of Shuttle-like, and presumably have to ride huge boosters into orbit like the old ones...
however
Private industry is hard at work with SSTO's, and at a fraction of the fuel consumption, that just may be a promising avenue...

There are two answers to this question.First,NASA has plans for a manned Mars flight in a few years.The second answer to this is a base on the Moon.I believe the second has to be the first for it to work.Their are alot of cool web sites that go into this.NASA,JPL,Hubble,etc. etc.
