Yes.
Find your hosts file (usually in c:\windows\system32\drivers\etc ) and add the following lines to it:
127.0.0.1 domain.com
127.0.0.1 www.domain.com
where "domain" is the URL of the website you want to block. You may need to restart your browser/disconnect and reconnect to the internet before this works.

In internet explorer, click hte tools menu. Hit internet options, and select the privacy tab. go to the bottom where it says websites and hit edit. In the box that pops up eneter the URL and hit block or allow!!! Pretty simple. Also you can do it through your router if you have one!Should work with Mozilla too!
EDIT.. what rat_the_grum... will just redirect the URL to localhost (127.0.0.1).. doesnt really block it.

Tools -> Internet Options ->content,
"use the CONTENT ADVIDSOR" section
Click ENABLE,
Click on Approved Sites,
Enter blocked website,
Click never,
OK,
OK

I use System Mechanic 6 program. It has a program that allows me to choose any web site I want to block. It also allows me to block everything except specific sites.
