As the answers above specify, it's from the book by Douglas Adams "The Hitchhiker's Guide to the Galaxy". The story underlying this answer (42) goes approximately like that: once "a race of hyperintelligent pandimensional beings" decided to find out the answer to the question about life, the universe and everything. For this purpose they built a powerful computer which they called Deep Thought. But the computer had to think about the answer for seven and half million years. Meanwhile the people were waiting. So when the time was due, they came to Deep Though again and asked the Question again. Everyone was impatient to know the Ultimate Answer. And the super computer had it and it was... forty-two! The problem was, that the Computer, and, according to it, the askers themselves didn't see what the question actually was. So to understand the answer, people had to find the question now. I strongly advise to read the book - it's a real pleasure. And then when another person answers your question of the meaning of life as "42", you'll get it.

It comes from a book. Some guy decided to be a smart-a$$ about it, and give the meaning of life a mathematical equation that came out to equal 43.
