Most likely natural rubber unless the tongue twister was invented by a DuPont engineer whilst commenting on the reuben sandwich he ordered at the cafeteria. In this case the bumper is probably made of neoprene.

It is supposed to be safe but I didn't take any chance's... With my daughter she always put everything in her mouth so when ever she would chew on something I would just throw it out, because I knew if I just took it away from her she would go back to it and chew again, It was very hard breaking that habit.... I really encouraged the pacifier, it's good for developing their pallit and helps keep other things out of their mouth's....

If you are worried that it may be harmful to your child.
I wouldn't worry to much, they are safe and I haven't heard of any child getting sick from them by either bitting on them on sitting on them.
