You can design the store in Dreamweaver, but you'll need a script to process your forms, in asp, php, perl, or whatever. There are solutions you can buy, or there are freeware ones out there. You will need to do at least a little modification to adapt any third-party scripts you get. To make it secure you'll need to purchase an SSL certificate, from Thawte, Verisign, or other.
If you don't want to bother with scripts, you can use something like coolcart.com to do the processing.
You'll also need a credit-card processing gateway service, a merchant card account (or at least PayPal), and a bank account to send it to.

I built one based on aspdotnetstorefront. You buy their code, then modify it to fit your needs. There are different levels depending on what you need. This will save you a lot of development time.
For security you will need to buy a SSL.
