If it is a low wall then dig a footing, lay a 6" bed of concrete with about 3" on each side of the blocks. Bed and lay the blocks on mortar and point up.
If the wall is going to be of any significant height (i.e. can fall on someone) or you are not sure of the ground conditions then get someone in.
If you do go for it make sure you use a level at all times (vertical and horizontal).

Lay a footer with concrete, then use string and a string level to make sure the wall is level from end to end. They have little blocks that the string hooks onto that also hook over the end of the bricks, or you can use poles at each end of the wall to tie the leveling string to. Sometimes a laser level can be used too.

Personally I would go to my local D.I.Y store and pick up some information leaflets on how to build that perfect wall... there are also many reference books that you can retrieve from your nearest lending library or reference library that may help you out a great deal. One thing that I would suggest is that you have to be completely sure of what you are doing before you attempt to build the wall or it may turn completely disastrous and you may have to pay someone to put it right. If you aren't sure ask someone to help you... Good Luck!!
