That depends on what you consider the first day.
The day after sex? NO.
The first day of your period? NO, because you could just be late.
The first day of pregnancy? NO, because you don't actually get pregnant the day you have sex, I think it's actually anywhere between 24-72 hours after sex. The egg is fertilized in the ovaries or your tubes, and then travels through the tubes to your uterus. Then it implants itself into your uterus. This takes time. After it's in your uterus, your body starts creating HcG, the pregnancy hormone. This can be detected by home pregnancy test (hpt) or a blood test. HcG multiplies rapidly (roughly 2x every day or so), so once there's implantation, a blood test can detect it.

You will not physically know that you are pregnant until you miss a period. You can possibly have early symptoms such as frequent urination and nausea. But these will not show up until the third or fourth week. The majority of the time it's longer than that.

not usually, you can usually tell after a week or two without a pregnancy test...with one, a good one, you can tell after about 5 days
