You get into a league that has an acceptable draft date and time for you.
You draft a team using current NFL players. At this point, you should have researched which players are most desirable to have on your team and which positions will grant you the most points (ex. RBs and WRs are higher priority than K, DEF, and even most TEs).
After you have drafted a team, you select your starting lineup for the first week against your opponent. Using the provided scoring system for your league, you try to pick who you believe will garner the most points for you in that week. The team with the most points wins the face-off that week. The second week, you go through the same thing, but against a different opponent in your league.
BYE weeks are in effect, which means that certain players will have one week off during the season. This is crucial since you eed to have backups from your bench cover for your players at least once ... it is important to draft well and keep an eye on the waiver wire for available players (to add while removing players that you feel are not helping you win the games).
Since this seems to be your first time playing, try to find a free league. That way, you only lose games and not anything else ... like an entry fee. If you don't like it, then at least you saved yourself a few bucks.
Check out NFL.com, Yahoo or ESPN for fantasy football leagues and read up on them to see if you like what you see from them.

There are several ways to play. Variations can be based on how points are allocated, how drafts are performed, if you use salary caps, playoff format, etc...
Basically you pick players, determine who you want to start that week, and add up the points earned by the players. Whoever has the most points wins.
