If you're talking about the longest running TV show, then it would be NBC's "Meet The Press", which first aired on November 6th, 1947. If you're talking about a fictional show, then that would be "Gunsmoke", which ran for 635 episodes from 1955 to 1975, and starred James Arness as Marshal Matt Dillon and Milburn Stone as Doc Adams.

Well, I would have to say one of the soap operas:
All My Children. Susan Lucci plays Erica Kane the notorious backstabbing sl-ut. And of course, you have David Canary who plays the notorious Adam Chandler.
The show first aired on January 5, 1970 as a 30 minute show. In 1977 it went to a one hour show. Of course, there are more actors that can be mentioned but I chose the top 2 that always has something going on in their lives.

"Meet The Press", if you include its time on radio.
Without taking the time to "look it up", do any research,
would guess
60 Minutes
But Bonanza
Gunsmoke
had long initial runs
And if you include re-runs,....I Love Lucy will probably beat all the records. Although Seinfeld looks like it could have a shot, eventually.
