For a single scene the #1 spot has to go to Airplane,when Jeff Daniels is pouring sweat in the pilots seat. For the movie with the most sweat I'd have to say it's Vision Quest.Although Mickey Rourke and Lisa Bonet certainly work up a sweat in the movie Angel Heart.

The general consensus is "Cool Hand Luke", but "Ben Hur" is up there as well. Also "Inherit the Wind" in a non-sexy, "let's hold a trial in the middle of a heat wave with no air conditioning" way.
