I agree with some of the answers that are on here already...
The Van Gogh Museum is fantastic, especially if you can go during a special exhibit. Anne Frank's House is really great. It's moving and sad, but important to see.
I also really love the Kukenhof (sp?) Gardens. The only problem is that you have to go in May because that's when all of the beautiful tulips bloom.
Just walking the streets of Amsterdam, crossing the canals, soaking in the culture is great. Oh, and if you like falefal, go to Maoz...it's sooo good!
One more tip, Amsterdam is a pretty safe city, but beware of pickpockets. Every time I go, I see someone getting robbed. You just have to pay attention and you'll be fine...

Anne Frank's house if you have a kid.
Amsterdam is so beautiful, unique, genuine and exotic that just being there is like being in a totally different world! the houses are so different from our culture, its history is sad, but plays a big part in our life.
Ride one of those water-buses. they would give you great info bits...like that amsterdam is actually made on water, or why everyone has a huge black bike. or why the houses are all so thin, small, and slanted?
the people there are especially welcoming. they speak great english and feel proud enough to give you a year's worth of history in a day. i'v only been there in winter, but that was still breathtaking (literally). but i guess all seasons are great.
i haven't answered your original question yet, and i honestly can't, but i just wanted to give you some pointers. ;)
PS: careful wid the bikes! don't disrespect their lanes because they don't have engines. the people ride em pretty fast, i almost go ran over. but notice how the entire city is made uniquely for bikes. get a ride...its sooo fun!!!!!

Going around the old canals on a boat on a nice warm summer day/night. You get to see the side of Amsterdam that you'd completely miss if you're on dry ground. It's just awesome going under those lit arched bridges, and when you're hungry just moor your boat on some cafe/restaurant that has piers exactly for this purpose and have a nice dinner.

The canals are nice! The heineken brewery is pretty interesting! (free beer) I think you should take a train or bus ride to the clogg place or rotterdam!

Quite frankly, when the trip is over, you won't remember anything about the city (wink. wink). Only thinkg to remember is respect the bikes. Don't step in the bike lanes when you're in your 'daze.' They'll run you over.
