If you mean western liberal democracy, fundamentalist Islam is a certainly a big threat. Effectively we're living during World War 3, except it's a protracted, undeclared war of incompatible ideologies. I don't think, however, that Islam is - or will ever be -capable of overturning the West as a power bloc, simpy because it's too reactionary to convert a majority to its cause. When the oil runs out, the Islamists will have much less literal and political currency from their spiritual homelands, will be more vulnerable to attack without bargaining power, and will be forced to rely entirely on piecemeal fundraising. Disregarding Palestine, It may be no bad end to all this when that happens and they are left to wage war on each other, with only a fragmented diaspora of believers vainly hoping for a global Muslim revolution that can never happen.

The single most threat to civilization would be asteriods.

Global warming is a slow process to wear off the civilization. Islam or other religions don't want to kill the people, they are not a threat to civilization. For those who think so go get the knowledge of history - Vietnam war, WW- II.

Apart from nuclear bombs and WMDs, no human made things can create destruction enough to wear off the civilization. It is nature who can create or destroy entrie civilization.

Forget about the Middle East, that area of the world has been in termoil since the beginning of time. How about GLOBAL WARMING, whats the weather been like in your neck of woods lately? Everyone blows it off as if it is only a concern of the excentric tree hugging folks. When infact it is a serious concern in the pentigon. People just don't pay attention, they are more concerned with paving over our forests, driving their gas guzzeling SUV's and poluting our water ways. When will people wake up and stop being so egocentric...........the world does not revolve around us. Your second runner up is our government. They want the world, one government, a "new secular order" (its right there in plain view every time you make a transaction with cash), watch out for them, or you'll be living in 1984.
