No it won't. Your body will replenish the lose of blood with making more blood. When the heart does that, the oxygen will be replaced as well, at the same time.

No it doesn't. And beside they only take blood when you are healthy and not suffering from diseases that could endanger your life. Your bone marrow produces RBC continuously, it's even healthy to donate blood as the old one gets replaced.
