Here are a few clues, but keep in mind that sometimes cultural differences can account for some of these behaviours as well.
1) Lack of eye contact
2) Looking down when speaking.
3) Head dropped when sitting, standing, walking.
4) Stuttering
5) Indecisiveness
6) Tenative movements (as opposed to purposeful ones)
7) Fidgety or nervous behaviour
I am sure there are many others, just a few i thought of offhand.

By their handshake. A firm handshake is the sign of self respect and self worth. A weak handshake shows lack of these traits. Looking at the floor during a conversation, or avoiding eye contact nervously will also be apparent. Making "fat jokes" or "stupid" comments about ones self is a pretty good sign, also.

Often people with low self esteem are great actors and actresses....They come off very confident and self assured but inside they are feeling just the opposite.

Well I can give you one example. Some people seem overly confident, but that's them just trying to pretend they are confident when they really have a low self-esteem. They are usually really conceited.

If you say they are pretty and they ignore you, or if their head is always down looking at the floor then that should give you a clue.
